dc.description.abstractWound infections are the major problems in our health care facilities. Pseudomonas aeruginosa is the most common pathogen causing wound infections and it is known to possess intrinsic resistance to many antibiotics. Objectives of this study were to; isolate and biochemically characterise Pseudomonas aeruginosa from wound samples, determine the antimicrobial susceptibility patterns of the isolates, Characterise the plasmid profile of the resistant isolates and determine risk factors associated with Pseudomonas aeruginosa in wound infections. One hundred and fifty (150) wound swab samples were collected from patients attending some hospitals in Zaria, Kaduna State Nigeria. The samples were cultured on cetrimide agar, twelve samples produced green colonies on the agar. The twelve isolates were subjected to biochemical(motility, Simmon citrate, oxidase, indole and triple sugar ion agar tests) and microscopic(Gram stain reaction) analysis. The twelve (12)isolates were positive for Pseudomonas aeruginosa. Antibiotic susceptibility test was carried out the twelve (12)isolates using eight different antibiotic discs(Ciprofloxacin, Tetracyclin, Chloramphenicol, Gentamycin, Ampicllin, Amoxicillin-clavulanate, Nitrofurantoin and Erythromycin). Only one (1) isolate was susceptible to the eight (8)antibiotics while the eleven (11) isolates were resistant to; Ampicllin, Amoxicillin-clavulanate, Nitrofurantoin and Erythromycin. Plasmid profile analysis was carried out on the twelve (12) isolates; no plasmid was isolated on the organism that was susceptible to all the antibiotics but in those organisms that were resistant to some antibiotics plasmids were isolated. On 0.8% agarose gel was isolated from twelve wound swab samples; some of the isolates were resistance toChloramphenicol and Erythromycinbut all isolates were completelyresistance toAmpicllin, Amoxicillin-clavulanate, and Nitrofurantoin. The isolates that were resistance to many antibiotics harbored plasmids with molecular size of 800bp. Risk factors associated with contamination of wound with Pseudomonas aeruginosa are age, sex and location of wound on the body.en_US
