dc.description.abstractThe extraction efficiency of continuous and batch extraction methods using four solvent systems of increasing polarities namely petroleum spirit 40 - 60 °C, Ethyl ether, 80% acetone and methanol were studied on dried rhizome of Cochlospermum tinctorium A. Rich. Phytochemical screening of extracts obtained by both extraction methods revealed the presence of alkaloids, tannins, saponins, cardiac glycosides, carbohydrates, flavonoids and anthraquinone glycosides which are reported for the first time. Preparative TLC of the ethyl acetate fraction with petroleum spirit (60 - 80 oC): ethyl acetate (3:2) yield extracts 1 - 6. Extracts 1 - 6 were characterized by RF value determination and reaction with spray reagents. UV spectra analysis shows extracts 1, 2, 5 and 6 to be flavanone glycosides and 3 and 4 to be flavonol glycosides. Cytotoxic test using Brine Shrimp Lethality assay was carried out on four crude extracts. The LC50 value for n-butanol, 80% acetone and methanol extracts were 437 ± 8 ug/ml, 240 ± 3 ug/ml and 871+5 ug/ml respectively. Meanwhile the ethyl acetate extract was lethal at all concentration level administered. Antimicrobial sensitivity tests of ethyl acetate, n-butanol, 80% acetone and methanol extract were carried out on Staphylococcus aureus, Pseudomonas aeruginosa, Klebsiella pneumoniae and Esherichia coli.
