Abstract
ABSTRACT
Fifteen soil samples obtained from the garden, refuse dump and
grassland in Ahmadu Bello University, Zaria, were screened for
Streptomyces spp. On the whole, 62 isolates of Streptomyces spp were
obtained. Characterization of these isolates using the "ISP Criteria"
gave 28 species of Streptomyces. 3.2% of the total isolates could not be
characterized. Morphological and physiological characterization of the
isolates based on the "ISP Criteria" showed that, the main coloration
found in all the isolates were gray, yellow and white with the occurrence
of 86.7%, 8.3% and 5.0%, respectively. The spore chain morphology was
rectus-flexibilis (RF) for 22 isolates (36.7%) and spira (S) for 38
isolates (63.3%). All the isolates had smooth spore surfaces. Only 23.3%
of the isolates were chromogenic. 76.7% of the isolates did not show any
chromogenic reaction on any of the three test media. 70% of the isolates
produced hydrogen sulphide, from either Peptone Iron Agar or Lead Acetate
Agar. The carbon utilization test revealed that the isolates can utilize
between 5-11 carbon sources. Based on the "ISP Criteria", 8 groups of
organisms were obtained. The percentage of occurence of Group I to VIII
are 6.4%, 24.2%, 16.1%, 37.0%, 1.6%, 4.8%, 1.6% and 4.8% respectively.
Group IV had the highest percentage of occurrence. 32.3% of the total
isolates were found to have antibiotic activity made up of 16.1% of
isolates have antibacterial activity, 9.7% antifungal activity and 6.5%
broad spectrum activity.
