Abstract
Alpha glucosidase inhibitors regulate postprandial hyperglycemia (PPHG) by impeding the rate of carbohydrate digestion in the small intestine and thereby hampering the diet associated acute glucose excursion. In this study, in vitro and in vivo alpha glucosidase inhibitory effects of the leaf extract of Carica papayalinnwere evaluated. The dried powdered leaf of C. papaya was extracted for 48 hours using four different solvents (water, methanol, ethyl acetate and n-hexane), after which the extracts were subjected to in vitro alpha glucosidase inhibitory studies.Methanolic extract being the most activewith an IC50 value of 171.83 ± 26.89 μg/ml was subjected to fractionation via column chromatography separation and six fractions were obtained which were subjected to further in vitro alpha glucosidase inhibitory studies.Fraction A (FA) had the lowest IC50(78.62 ± 25.23 μg/ml) than the other fractions and even lower than the standard drug acarbose (161.08 ± 41.01). Therefore, FA was selected for the in vivo maltose and sucrose tolerance tests as well asgas chromatography - mass spectrometry(GC -MS) analysis. The in vivo tests showed that FA had active inhibitory effects on maltose tolerance within the first 30 mins and on sucrose tolerance within the first 60 mins. The computed area under the curves (AUC (0 – 120)) further indicate that FA (at 200 mg/kg bw) have better inhibitory activity on maltase than sucrase. The GC - MSanalysis of FA indicates the presence of phenolics, various fatty acids and their derivatives. It was concluded that the leaf extract of C. papaya possess alpha glucosidase inhibitory effects and may provide a natural and safe remedy for the reduction of postprandial hyperglycemia which will therefore help to reduce the risk of developing metabolic disorders
