Abstract
ABSTRACT
Two Aspergill us species, (Aspergi 11us fumigat us- SSC36 and A s p e r g i l l u s n i g e r - C S 1 4 ) with enhanced cellulase
production were isolated from soil and decayed plant materials. These organisms were used to study the rate of cellulase production using cellulosic agricultural wastes {wheat straw and rice husk) as substrat. Combined alkaline and steam pretreatments to delignify the substrat enhanced cellulase production by the
organisms. The effects of different nitrogen sources on the activity of the enzymes were studies. The cellulase produced by Aspergi11 us fumigatus-SSC 35, grown at 28°C with an initial pH of 5.0 had optimum activity at pH 3.0 and 30°C in wheat straw and rice husk media. The enzyme was unstable above 30°C. The cellulase produced by A s p e r g i l l u s n i g e r CS1 4 grown at 28°C with an initial pH of 5.0 had optimum activity at pH 5.0 - 6.0 and 25°C in wheat straw medium while optimum activity at pH 4.0 and 30°C was recorded in rice husk medium. The enzyme was unstable above 25°C and 30°C in wheat straw and rice husk media respective1y.
FOR ASpergilllus fumigatus-SSC36, 1.0% (W?V) of groundnut cake (kuli-kuli) and 0.5% (w/v) of peptone (oxoid) promoted the highest enzyme production in wheat straw and rice husk media respectively. Enzyme production
was highest for A s p e r g i l l u s n i g e r CS1 4 using 1.0% (W/v) Urea and 1.0% (w/v) peptone (oxoid) in wheat straw and
rice husk media respectively. The results of this study highlight the industrial
potentials of these cellulosic materials as substrates for cellulase production.
