Abstract
A s i n g l e s o l v e n t system was developed under
t h e same c o n d i t i o n s for the a n a l y s i s of p a r a c e t a m o l,
c h l o r o q u i n e and promethazine in some p h a r m a c e u t i c al
p r e p a r a t i o n s (Maxiquine and Daga-Quine syrups, and
Daga-Quine t a b l e t s ) using High Performance Liquid
Chromatography (HPLC) Waters Model 204 f i t t e d with
U.V. d e t e c t o r Model 441 with a fixed wavelength
(254nm) and Model U6K s e p t u m l e s s i n j e c t o r . A pump
Model 510 at a flow r a t e of 1.5ml/min, with a
p r e s s u r e of 2000 PSI. The column used was a uBonda-
Pak (Radial-Pak) 0-18 column (l5cm x 3.9mm I . D . ) with
a mobile phase c o n s i s t i n g of a c e t o n i t r i l e : s o d i um
dihydrogen phosphate : t r i e t h y l a m i n e ( 6 0 : 3 9 : l ),
pH 7.2 at an ambient t e m p e r a t u r e . The r e t e n t i on
times (tR) of p a r a c e t a m o l , chloroquine and promethazine
were 3 . 0 , 5.8, and 12.0min. r e s p e c t i v e l y , while t h at
of the i n t e r n a l s t a n d a r d s were 4.2min for a c e t o a c e t a -
n i l i d e and 8.0 min. for p a p a v e r i n e , r e s p e c t i v e l y.
The e x t r a c t i o n r e c o v e r i e s were 97% for p a r a c e t a m o l,
and 90% for c h l o r o q u i n e and promethazine. Calibration
curves were l i n e a r from 20 to 100 ug/ml (r = 0.990)
f o r p a r a c e t a m o l , and 100 to 500 ug/ml (r = 0.999 and
0.990) for chloroquine and promethazine r e s p e c t i v e l y.
The Maxiquine syrup contained 96%, 95% and 101%
of p a r a c e t a m o l , chloroquine and promethazine r e s p e c t i v e l y.
The Daga-Quine syrup contained 96.7% and 96% of
paracetamol and chloroquine r e s p e c t i v e l y , while the
Daga-Quine t a b l e t s contained 101% and 103% of paracetamol
and chloroquine r e s p e c t i v e l y . The precision of the
method was 1.7% (n = 5) within run and 1.9% (n = 5)
for day-to-day run.
Spectrophotometric method employed for the
determination of chloroquine, paracetamol and promethazine
in Maxiquine syrup gave comparable r e s u l t s with the HPLC
method (100%, 100% and 97% of chloroquine, paracetamol
and promethazine r e s p e c t i v e l y ).
