Abstract
The anti-mycobacterial activity of Ficus sycomorus (stem bark, root, leaves and fruits) was studied in vitro against standard Mycobacterium tuberculosis strains using standard Nitrate Reductase Assay techniques with isoniazid and rifampicin as controls. The results revealed that the n-hexane fruit extract had activity against all the tested Mycobacterium tuberculosis strains, the activity was observed as follows: 400, 200, and 100 μg/mL against susceptible strain, 400μg/mL against isoniazid resistant (R-INH) and rifampicin resistant (R-RIF) strains. The n-hexane leaves, stem bark and root extracts had no activity and also the methanol and aqueous extracts of fruit, stem bark, root, and leaves of Ficus sycomorus showed no activity against all the tested Mycobacterium tuberculosis strains. The n-hexane fruit extract were subjected to constitutive phytochemical analysis of which saponins and alkaloids had the highest percentage content of, (16.67±0.04 %) and (6.00±0.02 %) respectively. The n-hexane fruit extract was then partially fractionated using thin layer chromatography and Colum chromatography where four fractions A-D were obtained, of which B-D were more active than A. The minimum inhibitory concentration were determined from fraction B-D where fraction B had the lowest minimum inhibitory concentration against all the tested Mycobacterium tuberculosis strains. The minimum inhibitory concentrations were observed at 6.25μg/mL against susceptible strain and R-RIF, and 100μg/mL against R-INH. Therefore, these findings indicate that Ficus sycomorus could be used as herbal drug not only against Mycobacterium tuberculosis but also against Multi-Drug resistant tuberculosis.
