Abstract
Aqueous, Methanol, Ethyl acetate and Hexane roots extracts of Ziziphus abyssinica were investigated for their effects on gastric lesion in rats administered with1ml of 95% ethanol and the antiulcer activity of Ziziphus abyssinica was compared to that of control. Pre-treatment of animals with the extracts (300mg/kg and 600mg/kg) orally once daily for two weeks significantly (p<0.05) reduced the formation of ulcers induced by ethanol, with aqueous extract having the highest effect. Percentage inhibition was 95% and 93%. The hexane extract afforded the least protection with 23.8% and 28.6%. The aqueous extract was further fractionated with hexane, ethyl acetate and butanol. The yields were 0g, 0.76g, 21.3g respectively and 37.4g for the remaining aqueous fractions. The aqueous fractions were found to significantly reduced ulcer index, percentage inhibition being 52.6 and 57.1% while the butanol has 28.6 and 18.1% respectively. The volume of the gastric juice and gastric acidity was significantly decreased as compared to control group, with significant increase in pH. Also, glutathione concentration, superoxide dismutase and catalase activities were maintained in Z. abyssinica root fractions administered animals. From the column chromatography, four fractions were obtained and each was tested for antiulcer effect. Fraction C showed highest antiulcer effect. The results indicate that Ziziphus abyssinica root extracts possess antiulcer effect with aqueous extract and aqueous fractions having the highest protective effect. The gastro-protective effect of Z. abyssinica roots could therefore be due to its cytoprotective (increased mucus secretion), antisecretory (decreased gastric juice volume, decreased acidity levels and increased pH) and antioxidant properties.
