dc.description.abstractThe study evaluated the larvicidal effect of Crude Methanolic extract of Jatrophacurcas seed oil extract using Soxhlet apparatus on Rhipicephalus (Boophilus) decoloratusticks. Different concentrations of 6.25, 12.5, 25 and 50 mg/ml were prepared from a stock (100 mg/ml) solution of the sample, control of 1% tween 80 and standard acaricide, Amitraz (positive control) were used. The mortalities were observed at time intervals. The experiment was performed in triplicate. The extract had an LC50 and LC90of 2.88mg/ml and 6.76mg/ml respectively after 48h. Carbohydrates, cardiac glycosides, saponins, tannins, steroids and terpenes, alkaloids and flavonoids were detected. The extract exhibited a significant mean percentage mortality from the control across all the concentrations after 24h. The result confirms that J. curcas is a good alternative to be used as a larvicidal agent in the control ofRhipicephalus (Boophilus) decoloratus. Five groups of two (2) rabbits each were used to carry out haematologic and blood chemistry and histotological assessments of the effect(s) of graded levels of J. curcasmethanolic seed extract topically applied on the skin of the rabbits for 2 weeks. 0.2mls of blood was collected from each rabbit on days 0, 7 and 14. Serum biochemical results of Mean AST, ALT and TP showed no significant changes compared with the control except for ALP values. Mean haematological parameters obtained (PCV, Hb, WBC and RBC) did not show any significant difference from the control and also falling within normal recommended ranges. Histopathological findings of the skin after the application of the pure crude revealed hyperkeratosis of the epidermis, infiltration of inflammatory cells and necrotic hair follicles. The splenic vessels were congested with widened red pulp. The liver showed distended sinusoid and congestion of the hepatic vessels with aggregation of inflammatory cells in the sinusoids. Haemosiderosis was seen with the 10% crude extract. This study reveals that the pure methanol extract of J. curcas seeds exhibited some toxic effect on the skin with prolonged use, but generally at lower concentrations, it remarkably has mild effect because histopathologic finding showed no observable adverse effect. Biochemical and haematological evaluations showed mild effect or variation from the normal.en_US
